If it was only religious beliefs. A lot, if not most, are homophobic without ever thinking about their "religion".
The vast majority of homophobia has religious roots though. Even if the current iteration the person isexpressing isn't explicitly so. It'll be some childhood bull shit like their religious parents telling them Para-Norman is an evil movie, and they don't even realize its the cause later in life.
Religious roots or religious "justification"? Like they don't seem to put as much importance on other things their religion says is important, like the commandments about lying or being envious of their neighbours or most of what their religion's namesake said, who was pretty clear that punishing sinners isn't supposed to be a duty of his followers.

It's essentially just a single line and an out of nowhere head turn over perceived stereotypes. The entire movie does a lot of that actually.
